나프탈렌 분해세균의 분리와 NAH플라스미드의 naphthalene dioxygenase에 의한 indigo 생성

Abstract
나프탈렌을 분해하는 6균주를 분리하였으며 이들에 의해 방향족 탄화수소화합물인 나프탈렌, salicylate, catechol 등이 분해됨을 알 수 있었다. Catechol 1,2-dioxygenase와 catechol 2,3-dioxygenase의 활성을 측정한 바 이들 분리주들은 catechol에서 meta-cleavage pathway를 거쳐서 분해되는 것으로 추정된다. 분리주 Pseudomonas chlororaphis SME6의 95 kb의 NAH플라스미드와 P. fluorescens SME11의 64 kb의 플라스미드가 pRK2013 helper plasmid에 의한 triparental mating에 의하여 전달되었으며, 이들 플라스미드 상에 nah유전자가 존재함을 확인하였다. 또한 방향족 탄화수소 중 m-MBA (methyl-benzylalcohol), naphthalene, benzoate, salicylate에 의하여 naphthalene dioxygenase가 유도 되었으며, 그 중 m-MBA가 가장 우수한 유도물질임을 indigo를 정량함으로써 알 수 있었다.
Six strains for the degradation of naphthalene were isolated and they utilized naphthalene, salicylate, catechol. and benzoate as aromatic hydrocarbons. Naphthalene was degraded by Pseudomonas chlororaphis SME6 or P. fluorescens SME11 through meta-cleavage pathway. NAH plasmids, 95 kb from SME6 and 64 kb from SME11 were transfered to P. putida UWC6 with aid of helper plasmid pRK2013. Transconjugants were able to utilize naphthalene, showing NAH plasmids carries nah gene clusters and produce indigo from indole using naphthalene dioxygenase induced with succinate. Among aromatic hydrocarbons, m-MBA, naphthalene, benzoate, and salicylate induced naphthalene dioxygenase; m-MBA was identified as the best inducer by quantitative assay of indigo product.
