In the two-dimensional electrophoresis images, the existing methods are focused on automation, so that they are very fast, and convenient but have poor accuracy in detecting background-like or abnormally-shaped spots. In this paper, we propose three new approaches to make up for these weak points of the traditional methods. First, researchers can customize the height of spots in order to detect small and feeble protein spots. Second, the protein spot is defined based on the average heights of spot peaks, not based on their shapes. Third, researchers can examine the shapes of spots and make an informative decision by visualizing spot shape data on the screen. The experiment results show that the accuracy of spot detection is improved by trying not to miss very fine and irregular spots which are ignored in the previously published papers and by allowing users to make subjective decisions about the range of spot heights and shape.
