The genetic polymorphism of cytochrome P450 (CYP) 2A6 has been emphasized to induce variations in smoking behavior, and in susceptibility to smoking-related cancer. However, there were a few studies concerning the CYP2A6 polymorphism in Korean populations. Therefore, focusing on presence of the CYP2A6 null allele (CYP2A6*4), I studied distribution of the CYP2A6 polymorphism in a Korean population (n=100) and its effects on sister chromatid exchange (SCE), a responsive biomarker for smoking or carcinogens. As results, the subjects who had the homozygous mutant type, the CYP2A6*4/*4, occupied 5.4% of the all subjects and most of them belonged to nonsmokers. However, the CYP2A6 polymorphism was not associated with the SCE levels (p=0.25), which were related to sex, smoking, and alcohol drinking. Therefore, presence of the CYP2A6 null allele may influence smoking behavior, however, it may be not involved in SCE-medicated cancer process. (Cancer Prev Res 12, 75-78, 2007)
