Galectin-3 (Gal-3), a member of the beta-galactoside-binding lectin family and expressed by many cell types including immune cells such as macrophages and numerous cancer cells, has been reported to be involved in colon cancer development and overexpressed in colorectal tumors. In order to determine relationship between inflammation and colon carcinogenesis, conditioned media (CM) from LPS-stimulated or non-stimulated macrophage, 3T3-L1 pre-adipocytes and differentiated adipocytes were utilized to induce inflammation in rat intestinal IEC-6 cells, followed by analysis of Gal-3 by western blot. Although preadipocyte and adipocyte cells showed increased secretion of tumor necrosis factor-α by incubation in the conditioned media from preadipocyte and adipocyte cultures, their culture media failed to regulate Gal-3 expression in IEC-6 cells. In conclusion, Gal-3 expression in IEC-6 cells was not affected by conditioned media collected from adipocyte and macrophage cultures, suggesting that cytokines produced from preadipocyte and adipocyte may not be directly involved in Gal-3 expression in rat intestinal IEC-6cells.
