Nanomedicine leverages nanomaterials and nanotechnologies to improve diagnosis and therapy by exploiting unique properties that arise at the nanoscale. Metal-organic frameworks (MOFs), composed of metal clusters and organic linkers, have gained increasing attention in nanomedicine. Among them, titanium-based MOFs (TiMOFs) stand out for their unique properties including high structural stability, tunable functionality, and low toxicity at appropriate concentrations. In this review, we highlight recent advances in Ti-MOF-based platforms for nanomedicine. We first introduce synthesis strategies of Ti-MOFs highlighting key characteristic features of each method, including synthesis process, reaction time, particle size, productivity, and reproducibility. Recent advances in Ti-MOFs for nanomedicine are discussed leveraging their unique photocatalytic activity, controlled drug delivery capacity, and anti-microbial property to enhance tissue regeneration. Subsequently, we discuss redox-, pH-, and light-responsive drug release, and photo-, sono-, and microwave-dynamic therapy for enhanced cancer treatment. Together, we demonstrate the great potential of Ti-MOFs as effective multifunctional nanomedicine platforms.
